I'll take Ledger's Joker any day of the week as the best of the bunch. He dominated every scene he was in. Given any number of re-watchings, you just can't take your eyes off the guy when he's on screen.
But, to be fair to both Nicholson and Romero, they were simply asked to do different things, and I think they both served the role well. As JE noted, while Christopher Nolan obviously took the Frank Miller "Dark Knight" and "Dark Knight Returns" series as his inspiration, Burton applied his dark-quirky-comic shtick to the series (full disclosure: I've never gotten into Burton's films...just not my cup of tea), and Romero, of course, acted in the overtly comic TV series. I thought Nicholson was able to play funny-but-dangerous quite well and really improved that movie from what it otherwise would have been, while Romero was great as a comic villain.
Next question, of course: Best Catwoman? That's a much tougher question, in my view. Julie Newmar? Eartha Kitt? Michelle Pfeiffer? At least we can rule out Halle Berry...
